AML1-ETO driven acute leukemia: insights into pathogenesis and potential therapeutic approaches

Megan A. Hatlen, Lan Wang, Stephen D. Nimer

Frontiers of Medicine 2012, Volume 6, Issue 3,   Pages 248-262 doi: 10.1007/s11684-012-0206-6

Abstract:

The AML1-ETO fusion transcription factor is generated by the t(8;21) translocation, which is present in approximately 4%–12% of adult and 12%–30% of pediatric acute myeloid leukemia (AML) patients. Both human and mouse models of AML have demonstrated that AML1-ETO is insufficient for leukemogenesis in the absence of secondary events. In this review, we discuss the pathogenetic insights that have been gained from identifying the various events that can cooperate with AML1-ETO to induce AML in vivo. We also discuss potential therapeutic strategies for t(8;21) positive AML that involve targeting the fusion protein itself, the proteins that bind to it, or the genes that it regulates. Recently published studies suggest that a targeted therapy for t(8;21) positive AML is feasible and may be coming sometime soon.

Abstract: Here we review the recent advances in the biosynthetic pathway of terpenoid indole alkaloids (TIAs) inSecologanin is derived from terpenoid (isoprenoid) biosynthetic pathway, while tryptamine is derivedfrom indole biosynthetic pathway.Although many genes and corresponding enzymes have been characterized in this pathway, our knowledgeon the whole TIA biosynthetic pathway still remains largely unknown up to date.

Human menstrual blood-derived stem cells alleviate autoimmune hepatitis via JNK/MAPK signaling pathway

Frontiers of Medicine 2023, Volume 17, Issue 3,   Pages 534-548 doi: 10.1007/s11684-022-0953-y

Abstract: Autoimmune hepatitis (AIH) is a severe globally distributed liver disease that could occur at any age. Human menstrual blood-derived stem cells (MenSCs) have shown therapeutic effect in acute lung injury and liver failure. However, their role in the curative effect of AIH remains unclear. Here, a classic AIH mouse model was constructed through intravenous injection with concanavalin A (Con A). MenSCs were intravenously injected while Con A injection in the treatment groups. The results showed that the mortality by Con A injection was significantly decreased by MenSCs treatment and liver function tests and histological analysis were also ameliorated. The results of phosphoproteomic analysis and RNA-seq revealed that MenSCs improved AIH, mainly by apoptosis and c-Jun N-terminal kinase/mitogen-activated protein signaling pathways. Apoptosis analysis demonstrated that the protein expression of cleaved caspase 3 was increased by Con A injection and reduced by MenSCs transplantation, consistent with the TUNEL staining results. An AML12 co-culture system and JNK inhibitor (SP600125) were used to verify the JNK/MAPK and apoptosis signaling pathways. These findings suggested that MenSCs could be a promising strategy for AIH.
